Matinas BioPharma, a clinical-stage biopharmaceutical company, has recently announced that it has successfully reached an agreement with the U.S. Food and Drug Administration (FDA) for a single Phase 3 registration trial to support the New Drug Application (NDA) for its drug candidate MAT2203. This agreement is a significant milestone in the development of MAT2203 for the treatment of invasive aspergillosis.
The ORALTO Phase 3 trial will focus on the desired initial target indication of early oral step-down treatment of invasive aspergillosis infections in adults who have limited or no treatment options. Invasive aspergillosis is a severe fungal infection that primarily affects immunocompromised individuals, such as those with weakened immune systems due to chemotherapy or organ transplantation.
MAT2203 is an oral formulation of amphotericin B, a well-known antifungal agent. The drug is encapsulated within Matinas’ proprietary lipid nano-crystal (LNC) platform, which enhances its bioavailability and allows for targeted delivery to the site of infection. The LNC platform also helps to minimize the toxicity commonly associated with traditional formulations of amphotericin B, making it a more tolerable treatment option for patients.
The agreement with the FDA for a single Phase 3 trial is a favorable outcome for Matinas BioPharma. Typically, drug candidates for life-threatening conditions like invasive aspergillosis require multiple trials for regulatory approval. However, the agreement allows for a streamlined development process, potentially accelerating the availability of MAT2203 to patients in need.
In previous Phase 2 studies, MAT2203 demonstrated positive results, showing efficacy and safety in the treatment of diverse fungal infections. The drug also exhibited a favorable pharmacokinetic profile, with high drug concentrations achieved in target organs such as the lungs.
The successful agreement with the FDA for a single Phase 3 trial reflects the potential of MAT2203 as a promising treatment option for invasive aspergillosis. Matinas BioPharma continues to advance the development and clinical evaluation of MAT2203, with the goal of providing patients with limited or no treatment options a much-needed solution for this life-threatening condition.
