Methods’

The Phase 1 dose escalation trial of BDTX-1535 included patients with recurrent GBM who had previously received standard therapies. The primary s were to assess the safety and tolerability of BDTX-1535 and to determine the maximum tolerated dose (MTD). Secondary s included evaluating the pharmacokinetics (PK), pharmacodynamics (PD), and preliminary anti-tumor activity of the drug. Patients received escalating doses of BDTX-1535 until the MTD was established, with close monitoring for adverse events (AEs) and response to treatment.

Results’

The trial enrolled a diverse cohort of patients with molecularly characterized GBM subtypes. BDTX-1535 demonstrated a favorable safety and tolerability profile, with most adverse events being Grade 1 or 2 in severity and manageable with supportive care. No dose-limiting toxicities (DLTs) were observed, and the MTD was successfully identified.

Encouragingly, BDTX-1535 exhibited preliminary signs of anti-tumor activity. Several patients experienced partial responses (PRs), while others achieved stable disease (SD) lasting beyond six months. Notably, the duration of treatment was prolonged in patients showing clinical benefit, suggesting potential efficacy in controlling tumor progression.

Discussion’

The results from this Phase 1 trial underscore the potential of BDTX-1535 as a therapeutic option for patients with recurrent GBM. The favorable safety profile and preliminary anti-tumor activity observed in the study warrant further investigation in larger, controlled trials. Additionally, the prolonged duration of treatment in responsive patients highlights the importance of continuing to explore BDTX-1535’s efficacy in this challenging patient population.

Conclusion’

The clinical data presented by Black Diamond Therapeutics at the 2024 ASCO Annual Meeting provide a promising outlook for BDTX-1535 in the treatment of recurrent GBM. These findings pave the way for subsequent phases of clinical trials to further evaluate the drug’s therapeutic potential.
